![]() Since the early studies have suggested that metformin acts by inhibition of complex 1 in mitochondrial electron transport chain and subsequently activation of AMPK, AMPK-independent targets have also been reported. Actually, in the last decade we moved from a simple picture, that metformin acts via the liver 5′ AMP-activated protein kinase (AMPK), to a much more complex one, reflecting its various mechanisms in different cells and tissues. These mechanisms remain a topic of considerable debate. The mechanisms underlying metformin actions appear to be complex and responsible for the pleiotropic effects of metformin.
